Bei neutralem oder leicht basischem pH-Wert (7–9) greift es schnell lösungsmittelzugängliche Lysin-NH*2*-Gruppen und N-Termini an. Obwohl sie im Allgemeinen seltener verwendet wird als DSG, DSS und DSSeb, kann eine Proteinstrukturanalyse, die auf chemischer Vernetzung mit DSA basiert, zusätzliche Strukturinformationen generieren.
